| Description | Human PDE3A (GenBank Accession No. NM_000921), (aa484-1141) with N-terminal GST tag, expressed in a Baculovirus infected Sf9 cell expression system Recombinant protein corresponding to aa484-1141 of human PDE3A with a N-terminal GST tag, expressed in a Baculovirus infected Sf9 cell expression system. Accession No. NM_000921 Specific Activity: 1100pmol/min/ug. One unit is defined as the amount of enzyme that will convert 1pmole of 3′, 5′-cAMP to 5′-AMP per minute at 37°C. Assay Conditions: 10mM Tris-HCl, pH 7.4, 10mM MgCl2, 0.1mg/ml BSA, 0.05% Tween-20, 200uM cAMP and serial dilutions of P4072-02A at 37ºC for 20 minutes. Quantified by 5′-nucleotidase cleaving the 5′-AMP product and releasing the phosphate group which is detected by Malachite Green Reagent. Applications: Useful for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Other applications not tested. Recommended Dilutions: Optimal dilutions to be determined by the researcher. Storage and Stability: Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. |
