| Description | Ready-to-use pre-packed columns of 1ml or 5ml bead volume in PBS with 0.02% sodium azide ≥5mg Protein A/ml Sepharose beads. Protein A-Sepharose Columns are prepared by covalently coupling recombinant Protein A to 6% cross-linked Sepharose beads. Protein A is a genetically engineered protein containing five IgG-binding regions of native Protein A. The cell wall binding region, albumin binding region and other non-specific regions have been eliminated from the recombinant Protein A to ensure the maximum specific IgG binding. The coupling technique is optimized to give a higher binding capacity for IgG and minimum leaching of recombinant Protein A. The IgG binding capacity of Protein A-Sepharose Column is ≥ 16mg human or rabbit IgG per ml of wet beads. Protein A-Sepharose Columns display high chemical and physical stability as well as high flow rate, hydrophilicity, and high gel strength. It can be used for IgG purification and immunoprecipitation. Protein A is a highly stable surface receptor produced by Staphylococcus aureus, which is capable of binding the Fc portion of immunoglobulins, especially IgGs, from a large number of species (Boyle and Reis, 1987). Each protein A molecule can bind 2 molecules of IgG, allowing the formation of a precipitate (Sjöholm, 1975). Applications: • Purification of monoclonal and polyclonal antibodies from culture media, serum, ascites fluid or hybridoma supernatants. • Isolation of antibody/antigen complexes in immunoprecipitation experiments, since only the Fc region is involved in antibody binding and the Fab region is available for binding antigen. Protein A binds to most human and mouse IgG subclasses (e.g., human IgG1, IgG2, IgG4; mouse IgG1, IgG2a, IgG2b, IgG3). It also binds to total IgG from cow, guinea pig, hamster, horse, pig, and rabbit. Protein A has little affinity to chicken, goat, rat and sheep. Binding Capacity: ≥16mg human or rabbit IgG/ml Protein A-Sepharose. Flow Rate: ~2ml/min Calculations based on the time required to pass 18ml of water through 2ml settled beads (column diameter 1.5cm). Usage: Reusable for up to 10 times without significant loss of binding capacity. Storage and Stability: Slurry may be stored at 4°C. Do Not Freeze. |
